Pharmacology  Ponazuril is a triazine antiprotozoal compound  developed by the Bayer Corporation under the trade name Marquis. It was first approved by FDA in 2001 for the treatment of mammalian central nervous diseases caused by Neospora hughesi and Sarcocystis neurona such as Equine Protozoal Myeloencephalitis (EPM). Unlike the regular anticoccidal compounds such as toltrazuril, ponazuril can cross blood-brain barrier, directly kill N hughesi and S neurona in the brain and spinal cord fluid, and reduce the clinical signs. Studies indicated that while killing coccidia, ponazuril did not affect development of host immune response. As the immune response develops, the animal can gradually decrease the drug dependence. In addition, by crossing placenta barrier, ponazuril can directly kill  N caninum which causes severe injuries in fetus. In vitro studies showed that ponazuril interfere with Neospora trophozoite division severely by increasing the amount of tubule mitochondria and reticulum, expanded the space around the nucleus. Ponazuril is therefore called coccidiocidal rather than coccidiostatic. In a study with dairy cows experimentally infected with N canimum by Sandra et al. , the cows were divided into two groups on the second day after infection in which one group treated with ponazuril and the other not treated with any drugs.  The results showed that the cows treated with ponazuril became serologically negative, whereas all positive in untreated cows, indicating that  ponazuril effectively killed N canimum in dairy cows. Other studies showed that in addition to Neospora,  ponazuril offered significant cidal activity against Eimeria coccidian and Gondii toxoplasma.

Warning: Horses sometimes may get stomach upset or a rash around mouth in which the drug is administered.

Directions: Marquis15%W/W of ponazurilantiprotozoal oral paste for horse,one syringe barel contains 127 g of paste, in which one g contains 150 mg of ponazuril (15% W/W). Marquis can be orally administered, and  can be used against coccocidiosis in  avian and rabbits by adding in drinking water and feed.
